Heart failure (HF) is characterized by deterioration of the electrical and contractile function of the heart due to structural and functional remodeling, leading to the development of arrhythmias and increased sudden cardiac death risk. This remodeling process is not fully characterized due to the scarcity of human tissue and the difficulty of translating results from cellular or animal models. Cardiac slices are a novel and highly representative model for cardiovascular research and they can be produced from tru-cut biopsies or papillary muscles from patients undergoing cardiac surgery. SK channels are a type of calcium-activated potassium channels that do not play a relevant role in normal ventricular electrophysiology. However, it has been hypothesized that these channels become more relevant in pathologies such as HF. The aim of the project was to determine the presence and function of SK channels in human failing ventricles by using ventricular tissue slices of living donors with HF. Significant APD shortening was observed upon activation of the SK channels, which was not observed in the control group. These results contribute to an improved understanding of HF and can serve as a basis for the development of new diagnostic tools and therapies
